As for the mytharc, my current theory is that yes, Sam killing Lilith is what breaks the last seal, thus explaining why Lilith believes she won't survive this war. In order to have a war, she has to die, and she doesn't want to.
I've always thought Ruby's allegiances were questionable, because otherwise she's just a pusher, and not an interesting one. She is, I think, the person who told Dean that Lilith held his contract, and I see no evidence she was lying.
